I'd like to endmill route holes in a piece of 1/8 inch thick sheet glass as well as route the outer profile rectangle. How would I do this on my Iconic machine? The smallest holes would be 1/4 inch. Resulting piece would be up to 8-17 inches wide by up to 2-4 inches high. I assume I'd have to clamp it with rubber padded clamps on a very stable spoil. What kind of bit to use? Acrylic is not an option here.

I never milled glass. But I milled acryllic and there are special bits for it. Since you need absolute flatness I would go with MDF spoil sheet. And use duct tape to attach it. Just holes? then you are ok. If you need to cut out bigger pieces that could be tricky how to remove them while milling (you will need to use double side tape to stick it to.
For my acrylics I use double side tape since I need to cut out the pieces and keep them, can't have them flying around and go into the spindle way....

Just wondering if you ever tried to cut glass. My first instinct is "No Way". I don't think carbide can handle it. I may be wrong though. Seems like the glass would be too brittle and would not cut well. Just curious
